Primary Applications in Microscopy

• Masson’s Trichrome Staining: This is its most legendary use in pathology and anatomy. Aniline Blue serves as the final counterstain in trichrome protocols to visually differentiate tissues. It binds specifically to collagen (the main structural protein in connective tissue), turning it a brilliant, deep blue. This creates a sharp contrast against muscle fibers and cytoplasm (which stain red) and cell nuclei (which stain black/purple). This layout is vital for diagnosing medical conditions like liver cirrhosis or kidney fibrosis, where tracking excess scar tissue (collagen accumulation) is key.

• Plant Histology: In botany, Aniline Blue is paired with other stains to highlight plant structures. It features a high affinity for callose—a carbohydrate structure found in pollen tubes and around sieve plates in the plant’s phloem (the tissue that transports sugars)—helping researchers track plant reproductive and vascular development.

• Fluorescence Microscopy: Interestingly, Aniline Blue can function as a fluorochrome. When exposed to ultraviolet (UV) light, it fluoresces a bright yellow-green. Scientists use this property to detect fungal cell walls (which contain chitin and glucans) infecting plant or animal tissues.
