Salkowski Reagent is a specialized colorimetric indicator formulated primarily for the qualitative detection and quantitative estimation of Indole-3-acetic acid (IAA), the principal plant hormone of the auxin class. Chemical Composition & Specification

Unlike the classic Salkowski variant used for sterol analysis, this formulation is optimized for indole detection. It contains precisely measured concentrations of Iron (III) Chloride (FeCl3) dissolved in an acidic matrix (typically optimized with Perchloric Acid (HClO4) or Sulfuric Acid (H2SO4). The inclusion of ferric ions (Fe3+) acts as a highly specific oxidizing agent tailored to react with the indole ring structure.

Principle of the Assay

The assay relies on a specific colorimetric reaction where the Fe3+ ions complex with and oxidize the indole-3-acetic acid molecule under strongly acidic conditions. This chemical interaction induces a distinct chromatic shift, transforming the solution from a faint yellowish tint to a vibrant pink to reddish-pink color. The color intensity directly correlates with the concentration of IAA present in the sample.
