Sodium thioglycollate is used in bacteriology as a reducing agent in Fluid Thioglycollate Medium, a type of broth that establishes anaerobic conditions to promote the growth of oxygen-sensitive (anaerobic) bacteria. It works by reacting with dissolved oxygen, lowering the oxygen concentration within the medium. This creates a gradient, allowing for the simultaneous cultivation and differentiation of aerobic, facultative anaerobic, and obligate anaerobic bacteria based on their oxygen requirements. The concentration of oxygen at different levels of the broth can be visualized using a redox-sensitive dye. 
How it works in Thioglycollate Medium 
  • Reduces Oxygen:
    Sodium thioglycollate chemically binds with dissolved oxygen, creating an anaerobic environment at the bottom of the tube.
  • Creates an Oxygen Gradient:
    Oxygen diffusion from the top of the broth, combined with the oxygen-reducing action of thioglycollate, forms a range of oxygen levels from high at the surface to near-zero at the bottom.
  • Differentiates Bacteria:
    • Obligate aerobes(require oxygen) grow at the top of the tube.
    • Obligate anaerobes(oxygen-intolerant) grow at the bottom.
    • Facultative anaerobes(can grow with or without oxygen) grow in the upper regions where some oxygen is present.
  • Uses Redox Indicators:
    A redox dye, like resazurin, is often included in the medium. It turns pink in the presence of oxygen, providing a visual indicator of the oxygen concentration at different depths.
